N-timer™ Insulin

The reagent stains live/viable Islets of Langerhans in vitro (WITHOUT FIXATION, WITHOUT PERMEABILIZATION). N-timer™ molecules are small synthetic peptides with a highly specific binding region targeting insulin. The N-timer™ Insulin molecule crosses tissues and cellular membranes in live islets, and remain only in cells containing high levels of Insulin. 

N-timer™ Insulin (or Beta Cell Marker) is available in a number of variants.

The N-timer™ Insulin staining process is rapid, simple and reproducible. Each N-timer™ insulin carries one dye molecule and will transport the dye across biological membranes resulting in a simple and rapid staining process.

It showed no affect on the survival of Islets in culture for mouse or human tissue, It also did not affect functional status of the isolated islets (i.e. insulin release, metabolism of Islets).

IMPORTANT

Reconstitution and conjugation of the N-timer™ The N-timer™ should be dissolved in 10 μl of the buffer recommended (by the dye manufacturer) for coupling the dye reagent. This N-timer™ is soluble in water, DMSO and DMF. It is recommended that 4μg of dye, ONLY USE AMINE REACTIVE DYE CHEMISTRY WITH THIS REAGENT, is used in the coupling reaction (according to the dye manufacturers instructions). It is also recommended that highly specific reagents for the attachment to amines are use as there are other free and vital nucleophiles are present (sulphhydral); thus chemistries like Isothiocynates are not recommended.After coupling to the dye, the N-timer™ can be purified by reverse phase HPLC. Do not use size separation products that are designed for proteins or anti bodies as the small size of the N-timer™will not separate from the dye and the loss of N-timer will be significant.
